Sure! Make sure nothing else is selected (e.g click between two files),
then press and hold Ctrl key while single-clicking on the desired file.

Thanks for your reply! My only problem is that I'm using the "list view" of nautilus and not the "icon" view so I cannot click between two files. This is why I'd like to find a keyboard/mouse command that unselects all other files except the one being "clicked" on. Have you ever used the list view with a lot of files in a small nautilus window? There's no where to click in the window to unselect files that have been selected. Do you understand what I mean? But thanks!
