you have to pass the ethernet device (eth0 or eth1...) to the script

./domain.up eth0

and maybe try to run it with sudo.

resolvconf is really not _meant_ to be run by hand, and yes it would need to
be run by sudo (as it is normally invoked by, iirc, dhclient and has to have
privilege to modify /etc/resolv.conf).
