So, can FAT32 drives be defragmented under linux?
A sideways question: has anyone seen meaningful timings/efficiency changes for FAT32 when it becomes fragmented?

Surely with the speed of even only recently modern machines and drives - along with the drive and OS buffering - and any data read-ahead by the
application - the fact that a file might be a bit fragmented must be getting pretty irrelevant by now.
I guess it helps the HD live longer if you keep the drives defragmented. Data throughput also increases. I use Windows for gaming, and when the drive is fragmented, I notice that the games sometimes pause a bit in order to wait for the HD to deliver the data (sounds/textures/etc).
Since gaming is considered a "real-time task", you might not notice any difference with non real-time applications.
