No, there are GUI frameworks that don't run on X, like e.g. NCurses.
But many do since X is *the* GUI platform for GNU/Linux.
GTK+ is a so-called widget toolkit that is a convenient wrapper for
X painting routines (CMIIW).
wxWidgets is a cross-platform widget toolkit (that also works on
other operating systems). In GNU/Linux, IIRC it works /on top of/
GTK+.
