Yep, mount your Knoppix 5.0.1/2006-06-01 iso image via loop or LiveCD
and copy all its contents to your USB drive and install grub on the USB
drive; you need 700+Mb free space on your USB drive. I have tested this
on an IDE drive/partition; this makes both the booting and, or other
regular operations quite faster.
The kernel (linux) and initrd image (minirt.gz) can be found in
<wherever.you.mounted.iso.or.livecd>/boot/isolinux/
Ideas and, or hints from isolinux.cfg file in the above said directory
can be reasonably helpful while creating a menu.lst for your grub.
