If you use the ext3 filesystem (this is the default in most distros),
this should not be a problem.
From /usr/src/linux/Documentation/filesystems/ext2.txt:
Filesystem block size: 1kB 2kB 4kB 8kB
File size limit: 16GB 256GB 2048GB 2048GB
Filesystem size limit: 2047GB 8192GB 16384GB 32768GB
And 4 kB block size is the default. So you shouldn't have any problem
with making a partition of 250 GB.
Another question is if you really want a partition so large. Even with
much smaller disks, it is convenient to have at least /home on a
separate partition, so for example if at some moment you choose to
switch to another distro (or change your root filesystem type or
whatever), you can wipe / completely because all your personal data is
on /home which is a separate partition. I also tend to have a separate
partition for data which changes infrequently and which I mount as
read-only to avoid accidentally deleting something form there even if I
do something stupid as root.
