Is there an other rpm, up2date or yum process running?

ps -ef|grep rpm
ps -ef|grep up2date
ps -ef|grep yum

if one of these give a process name, try to kill it and see if your rpu -U mypacage.rpm will work.

If not show us the output of rpm -Uvv mypacage.rpm
